- BML-278 is a selective, cell-permeable activator of SIRT1, a NAD⁺-dependent deacetylase involved in regulating cellular stress responses, metabolism, and aging. With high specificity for SIRT1 over SIRT2 and SIRT3, BML-278 enhances SIRT1 activity at low micromolar concentrations. In neuroscience, SIRT1 activation is associated with neuroprotection, improved mitochondrial function, and reduced neuroinflammation. SIRT1 modulates key transcription factors such as PGC-1A Alpha and FOXO, which are involved in oxidative stress resistance and neuronal survival. BML-278 is increasingly used in models of Alzheimer's and Parkinson's disease to investigate therapeutic strategies targeting age-related cognitive decline and neurodegenerative mechanisms.
