- GCO-(GPO)10-GCOG is a selective glycoprotein VI (GpVI) agonist with a collagen-mimicking triple-helical structure; which binds to human YadA with an affinity of 0.3 μM. GCO-(GPO)10-GCOG exerts its activity by binding to the Ig-like ligand-binding domain of GpVI. GCO-(GPO)10-GCOG relies on the hydroxyproline content in the GPO triplet; and its activity is closely related to its structural form: the monomeric form only induces limited platelet protein tyrosine phosphorylation without triggering aggregation; while the cross-linked form can trigger strong phosphorylation; complete aggregation; and GpVI-mediated adhesion. GCO-(GPO)10-GCOG can be used to study platelet-collagen interactions and YadA-host protein interference; and is applied in thrombosis-related research[1][2].
