- ML323 binds to the complex of USP1-UAF1 resulting in the inhibition of USP1 deubiquitylase activity (Liang, Dexheimer et al. 2014). The IC50 is reported to be 76 nM against Ub-Rho110 and 174 nM against K63-linked diubiquitin. Inhibition is reversible and exhibits mixed-mode kinetics. ML323 showed little or no inhibitory activity against a collection of DUBs including USPs 2, 5, 7, 8, 10, 11, 14, and 21, UCH-L1 and UCH-L3, and SenP1.
