After spiking FLICA® into the cell culture media, the cells were treated with either a negative control (left) or nigericin (20 uM, no.6698) to induce caspase-1 activity (right) for 24 hours. Gasdermin D mediated pyroptosis in vivo and in vitro and correlated with the accumulation of DNA damage. NPCs were isolated from Gsdmd fl/flmice and GsdmdcKO-Nes mice and cultured in vitro for 24 h, and cells were stained with FLICA-CASP1 (Cat no 9122) for 1 h and Propidium iodide.
