- After fixation with paraformaldehyde; formalin or other aldehyde-based reagents; antigenic determinants become masked by aldehyde cross-links; carboxymethylation; and protein–protein cross-linking. These modifications alter protein conformation; reduce the number of accessible epitopes; and lead to false-negative results or weak staining in immunolabeling. The aldehyde-induced cross-links are reversible; under appropriate conditions (e.g.; high temperature or enzymatic digestion) the original protein conformation can be restored. This process is termed antigen retrieval (AR) and is a critical step in immunohistochemistry. Multiple retrieval methods are available; heat-induced epitope retrieval (HIER) or enzymatic retrieval can be selected according to tissue type and antigen characteristics. This product; Tris-EDTA Antigen Retrieval Buffer (10×; pH 9.0); is a concentrated buffer. Dilute 10-fold to obtain a working solution of pH 9.0 containing 1 mM EDTA; suitable for antigen retrieval.
