- After fixation with paraformaldehyde; formalin; or other aldehyde-based reagents; antigenic epitopes become masked by aldehyde- and carboxymethyl-mediated cross-links and by protein–protein cross-linking. These alterations distort the native protein conformation; reduce the number of accessible antibody-binding sites; and lead to false-negative results or diminished staining intensity. Because aldehyde-induced cross-links are reversible; the original conformation can be restored under high temperature or proteolytic conditions; this step is known as antigen retrieval (AR) and is critical for reliable immunohistochemistry. The optimal retrieval method—heat-induced epitope retrieval (HIER) or enzymatic digestion—should be selected according to tissue type. This product is a concentrated Tris-EDTA antigen retrieval buffer (20×; pH 9.0). Dilute 1:20 to obtain a working solution containing 1 mM EDTA at pH 9.0; suitable for antigen retrieval.
