- PAI-1 (plasminogen activator inhibitor-1) is a member of the serine protease inhibitor family. It is the main negative regulator of the fibrinolytic system, preventing plasminogen from converting into plasmin by inhibiting tissue-type plasminogen activator and urokinase-type plasminogen activator, thereby inhibiting fibrin degradation. It is mainly secreted by vascular endothelial cells, adipocytes, hepatocytes, platelets, and smooth muscle cells. The core function of PAI-1 is to regulate the balance between fibrinolysis and coagulation. However, its overexpression is associated with various diseases, including thrombotic disorders, fibrotic diseases, metabolic syndrome and obesity, and cancer.
