- Vascular cell adhesion molecule-1 (VCAM-1, CD106) is a type I membrane glycoprotein of the immunoglobulin superfamily. Mouse VCAM-1 has two isoforms: a 110 kDa transmembrane long isoform and a 45 kDa GPI-anchored short isoform, both releasable as soluble VCAM-1. VCAM-1 is induced on vascular endothelium by inflammatory stimuli and constitutively expressed on non-vascular cells including dendritic cells, macrophages, fibroblasts, bone marrow stromal cells, and others. It mediates cell adhesion and signal transduction by binding to ligands such as integrin A Alpha4B Beta1, A Alpha4B Beta7, A AlphaDB Beta2, and A Alpha9B Beta1. VCAM-1 is critical for leukocyte recruitment to inflammation sites, T/B cell development, hematopoietic progenitor cell homing, and may facilitate melanoma metastasis. Soluble VCAM-1 promotes angiogenesis and chemotaxis. It is also associated with pathological conditions like atherosclerosis, rheumatoid arthritis, systemic lupus erythematosus, inflammatory bowel disease, and allograft rejection.
