- Human coagulation factor IX is is a vitamin K-dependent plasma protein, composed of 416 amino acid residues that plays a crucial role in the intrinsic pathway of blood coagulation by converting factor X into its active form in the presence of Ca(2+) ions, phospholipids, and factor VIIIa. It is primarily synthesized in the liver and subsequently secreted into the plasma. During the intrinsic pathway of coagulation, activation of human coagulation factor IX releases a peptide segment, resulting in the formation of two polypeptide chains linked by disulfide bonds. The region that interacts with phospholipids resides in the light chain, while the catalytic active site of the enzyme is located in the heavy chain. Activated coagulation factor IX (IXa), in conjunction with activated coagulation factor VIII, activates coagulation factor X, facilitating the conversion of prothrombin to thrombin and thereby ensuring normal progression of the clotting process.Deficiencies in F9 are responsible for recessive X-linked hemophilia B (HEMB) , commonly referred to as Christmas disease. Notably, mutations at position 43 (Oxford-3, San Dimas) and position 46 (Cambridge) hinder propeptide cleavage, a mutation at position 93 (Alabama) likely disrupts binding to cell membranes, while mutations at positions 191 (Chapel-Hill) or 226 (Nagoya or Hilo) impede activation peptide cleavage.Additionally, defects in F9 can lead to thrombophilia associated with factor IX deficiency (THR-FIX) , which is characterized by an increased tendency toward thrombosis.
