PDC catalyzes pyruvate decarboxylate to acetaldehyde, which reacts under the action of ethanol dehydrogenase (ADH), and catalyzes NADH to convert into NAD+. NADH has a characteristic absorption peak at 340 nm. The PDC activity can be calculated by measuring the OD value at 340 nm.
