- Interleukin-2(IL-2), or T-cell growth factor(TCGF), is a 15-18 kDa glycosylated cytokine in the common gamma chain(G Gammac)family, with a short half-life under 30 minutes. It is produced by activated CD4+and CD8+T cells, G GammaD DeltaT cells, microglia, and hematopoietic stem cells.The IL-2 receptor consists of CD25(IL-2RA Alpha), IL-2RB Beta, and G Gammac subunits. IL-2 binding to CD25 recruits IL-2RB Beta and G Gammac to form a signaling complex. IL-2RB Beta is also used by IL-15, and G Gammac is shared by multiple cytokines including IL-4, IL-7, and IL-15.IL-2 drives T cell activation and expansion in vitro. In vivo, it is essential for regulatory T cell(Treg)development and function, preventing autoimmunity, but can also promote inflammatory responses via Th2 cytokines and trafficking gene regulation. In CD8+T cells, IL-2 enhances effector differentiation and memory formation.
