- Interleukin-15 (IL-15) is a pivotal immunoregulatory cytokine that structurally and functionally resembles IL-2. It is produced by various cells, including dendritic cells and monocytes. Its defining mechanism is trans-presentation: IL-15 binds with high affinity to its private receptor chain, IL-15RA Alpha, forming a complex presented on the cell surface. This complex then signals to adjacent NK and T cells via their IL-2/15RB Beta and common G Gammac (G Gammac) chains, driving survival and proliferation.IL-15 broadly activates both innate and adaptive immunity. It stimulates proliferation and differentiation of NK, NKT, CD8+ memory T, and B cells, promotes dendritic cell development, and activates neutrophils and macrophages to produce pro-inflammatory cytokines and chemokines (e.g., IL-8, CCL2). Intracellular signaling occurs via JAK1/JAK3, activating STAT3/STAT5 (and STAT6 in mast cells). Soluble IL-15RA Alpha, generated by shedding, acts as a natural inhibitor.Therapeutically, recombinant IL-15 is promising for cancer immunotherapy because, unlike IL-2, it expands cytotoxic lymphocytes without preferentially stimulating regulatory T cells (Tregs). Current strategies aim to overcome its toxicity by combining it with checkpoint inhibitors, cancer vaccines, or cytokines like IL-7 for adoptive cell therapy. It also plays a metabolic role by inhibiting lipid deposition in adipocytes.
