- Interferon-beta (IFN-B Beta), a type I interferon, plays a key role in innate immunity against infections, tumors and inflammatory stimuli. Produced by fibroblasts, dendritic cells, macrophages and endothelial cells, it exerts diverse functions via specific signaling pathways and is clinically used to treat multiple sclerosis (MS). IFN-B Beta mainly signals by binding to the heterodimeric receptor (high-affinity IFNAR2 and low-affinity IFNAR1), activating the Jak-STAT pathway. This regulates interferon-responsive genes encoding antiviral, proliferation-regulating and immunoregulatory proteins. It can also function via IFNAR1 alone, independent of Jak-STAT, enhancing its signaling versatility.Its biological effects include antiviral/antibacterial activities, regulation of B-cell development, myelopoiesis and LPS-inducible tumor necrosis factor. Notably, it is more potent than IFN-A Alpha in inducing apoptotic and antiproliferative pathways for tumor control, and protects dopaminergic neurons by maintaining neuronal homeostasis.
