- TIM-1 (T-cell immunoglobulin and mucin domain protein 1, also known as KIM-1 and HAVCR) is a 70-80 kDa type I transmembrane glycoprotein belonging to the TIM immunoglobulin superfamily (1-4). This family regulates the balance between Th1 and Th2 immune responses. Structurally, TIM proteins contain an N-terminal immunoglobulin variable (IgV) domain, a glycosylated mucin domain, a transmembrane region, and a cytoplasmic tail.TIM proteins recognize phosphatidylserine exposed on apoptotic cells via a metal-ion-dependent binding pocket within the IgV domain. Notably, mouse TIM-2 lacks this pocket and does not bind phosphatidylserine. TIM-1 also interacts with P-selectin (CD62P) with higher affinity than with E- or L-selectin, mediating T-cell trafficking and costimulation. Studies using a contact hypersensitivity model indicate that TIM-1-P-selectin interaction facilitates the migration of activated T cells to inflamed skin.
