- Interleukin-8 (IL-8), also termed GCP-1 or NAP-1, is an 8-9 kDa CXC chemokine capable of binding heparin. It is produced by diverse cell types such as monocytes, neutrophils, fibroblasts, keratinocytes, mast cells, smooth muscle cells, dendritic cells, alveolar type II cells, and endothelial cells. IL-8 signals through two G protein-coupled receptors, CXCR1 and CXCR2, which form constitutive homodimers and can transiently heterodimerize. CXCR2, activated at low IL-8 levels, primarily mediates chemotaxis and MMP-9 release, promoting leukocyte adhesion and migration. In contrast, CXCR1 responds to high IL-8 concentrations and triggers respiratory burst and phospholipase D2 activation, enhancing neutrophil antimicrobial responses. IL-8 activity can be inhibited by complex formation with Serpin A1/alpha-1 antitrypsin, which blocks CXCR1 interaction. Beyond inflammation, IL-8 contributes to angiogenesis, atherosclerosis, and cancer progression by inducing VEGF expression, supporting endothelial cell growth, and facilitating tumor invasiveness and metastasis.
