- IL-33 (Interleukin-33) is a cytokine belonging to the IL-1 family and is mainly expressed by epithelial cells, endothelial cells, and fibroblasts. It signals through the ST2 (IL-1RL1) receptor in complex with IL-1RAcP, activating downstream pathways such as NF-κB and MAPK to initiate type 2 immune responses. Functionally, IL-33 acts as an “alarmin” released upon tissue damage, promoting activation of Th2 cells, ILC2s, mast cells, and eosinophils, and inducing the production of IL-5 and IL-13. In *in vitro* cell and organoid models, IL-33 is commonly used to simulate tissue injury–associated inflammation and type 2 immune microenvironments, particularly in studies of allergy, asthma, and epithelial–immune interactions.
