- Transferrin is an important glycoprotein composed of two domains, each containing an iron-binding site—one at the N-terminus and one at the C-terminus. Each site can bind one Fe?? ion with the cooperative involvement of one HCO?? ion. Its primary role is to transport and regulate iron metabolism, delivering iron to cells in a safe manner that avoids the toxicity associated with free iron ions. Iron is an essential cofactor for haemoglobin synthesis, electron transport chain components (such as cytochromes), and DNA synthesis enzymes, making it particularly critical for highly proliferative cells like cancer cells and stem cells. As a key additive in serum-free culture media, transferrin can substitute for serum as an iron source, ensuring cell survival and growth.
