- IL-12 (Interleukin-12) is a key pro-inflammatory cytokine and a member of the IL-12 family, primarily produced by antigen-presenting cells such as dendritic cells, macrophages, and B cells. It is a heterodimeric protein (p70) composed of two subunits, p35 and p40, linked by a disulfide bond. IL-12 promotes Th1 cell differentiation by activating the STAT4 signaling pathway, driving the commitment of na?ve CD4+ T cells into Th1 cells and stimulating the secretion of pro-inflammatory cytokines such as IFN-γ. It also enhances the activity of NK and T cells by promoting the proliferation and cytotoxic function of NK cells and CTLs (cytotoxic T lymphocytes). Furthermore, IL-12 suppresses Th2 responses by inhibiting IL-4-mediated Th2 cell differentiation, thereby reinforcing cell-mediated immunity.
