- R-Spondin-1 (RSPO1) is a secreted protein belonging to the R-Spondin family and plays a key regulatory role in the Wnt/β-catenin signaling pathway. By binding to Wnt receptors LRP5/6 and transmembrane proteins LGR4/5/6, RSPO1 inhibits negative regulators of the Wnt pathway and reduces the degradation of Wnt receptors, thereby significantly enhancing Wnt/β-catenin signaling activity. Through potentiation of Wnt signaling, RSPO1 promotes cell proliferation, stemness maintenance, and tissue regeneration, making it particularly valuable in stem cell culture. In 3D organoid culture systems, RSPO1—when combined with factors such as EGF and Noggin—significantly improves organoid formation efficiency and long-term viability. In stem cell cultures, its use together with Wnt3a supports self-renewal and suppresses differentiation. In summary, R-Spondin-1 is a critical factor in stem cell and organoid culture systems. By modulating the Wnt signaling pathway, it facilitates cell proliferation and maintenance of stemness, and is widely applied in regenerative medicine and disease modeling research.
