- LIF (Leukemia Inhibitory Factor) is a pleiotropic cytokine belonging to the IL-6 family. As a secreted glycoprotein, it activates downstream signaling pathways by binding to the LIFR/gp130 receptor complex and plays a critical role in cell culture, particularly in stem cell maintenance. Its primary function is to sustain pluripotency in mouse embryonic stem cells (ESCs) and induced pluripotent stem cells (iPSCs). In other stem cell contexts, LIF promotes the survival and proliferation of neural stem cells and, in combination with factors such as SCF and IL-3, helps maintain the stemness of hematopoietic stem cells.
