- IL-18 (Interleukin-18) is a pleiotropic pro-inflammatory cytokine belonging to the IL-1 superfamily. It signals through a heterodimeric receptor composed of IL-18Rα (binding chain) and IL-18Rβ (signaling chain), leading to activation of NF-κB and MAPK pathways. IL-18 exerts immunomodulatory functions by promoting Th1 immune responses and, in synergy with IL-12, induces IFN-γ production in T cells and NK cells, thereby enhancing cell-mediated immunity (e.g., in anti-tumor or anti-infection responses). Additionally, it activates NK cells, boosting their cytotoxicity and cytokine secretion. Under specific conditions—such as in combination with IL-23—IL-18 can also promote Th17 differentiation and has been implicated in models of autoimmune diseases.
