- IL-10 (Interleukin-10) is an important anti-inflammatory cytokine primarily produced by immune cells—such as T cells, B cells, monocytes, macrophages, and dendritic cells—as well as certain non-immune cells including keratinocytes. In cell culture, IL-10 suppresses the production of pro-inflammatory cytokines (e.g., TNF-α, IL-1β, IL-6, and IL-12), downregulates M1 macrophage activation, and promotes polarization toward M2 (anti-inflammatory) macrophages. In immune regulation, IL-10 inhibits the proliferation and function of T cells—particularly Th1 and Th17 subsets—while enhancing the immunosuppressive activity of regulatory T cells (Tregs). In macrophage or dendritic cell cultures, IL-10 can be used to induce an anti-inflammatory phenotype and mimic an immune-tolerant microenvironment similar to that found in vivo.
