- IL-6 (Interleukin-6) is a pleiotropic cytokine secreted by various cell types, including T cells, B cells, macrophages, fibroblasts, and endothelial cells. It plays a critical role in immune regulation, inflammatory responses, hematopoiesis, metabolism, and tissue repair. Signaling is mediated through a complex composed of gp130 and the IL-6 receptor (IL-6R), activating pathways such as JAK/STAT, MAPK, and PI3K. In immune cell cultures—including T cells, B cells, and macrophages—IL-6 promotes Th17 cell differentiation (associated with autoimmunity), suppresses regulatory T cell (Treg) function, and stimulates B cell differentiation into plasma cells, thereby enhancing antibody production. In hematopoietic stem cell cultures, IL-6 acts synergistically with other cytokines (e.g., IL-3 and SCF) to promote the proliferation and differentiation of hematopoietic progenitors, particularly along the megakaryocyte and granulocyte lineages.
