- IL-4 (Interleukin-4) is a cytokine primarily secreted by Th2 cells, mast cells, and basophils. It functions by binding to receptor complexes such as IL-4Rα/γc or IL-4Rα/IL-13Rα1, activating the JAK-STAT6 signaling pathway to regulate immune responses, cell proliferation, and differentiation. IL-4 modulates immune cell differentiation and function by promoting Th2 cell polarization and is commonly used in?in vitro?B cell culture systems. In organoid cultures, IL-4 acts synergistically with IL-13 to drive goblet cell differentiation and modulate the immune microenvironment in intestinal organoids. Additionally, IL-4 can suppress neuronal differentiation and promote the generation of astrocytes.
