- Thrombopoietin (TPO) is a crucial hematopoietic growth factor belonging to the hematopoietic cytokine family. It serves as a central regulator of megakaryocyte development and platelet production, stimulating the differentiation of hematopoietic stem cells into the megakaryocytic lineage and ultimately leading to thrombopoiesis. Upon binding to its receptor c-Mpl, TPO activates signaling pathways such as JAK/STAT and PI3K/AKT, promoting the survival, proliferation, and long-term self-renewal of hematopoietic stem cells. When used in combination with other cytokines—such as SCF, IL-3, and IL-6—TPO enhances the expansion efficiency of hematopoietic stem and progenitor cells.
