- IL-7 (Interleukin-7) is a cytokine produced by bone marrow stromal cells, thymic epithelial cells, and other cell types. It belongs to the four-helix bundle cytokine family and exerts its functions primarily by binding to the IL-7 receptor, thereby activating signaling pathways such as JAK-STAT, PI3K-AKT, and MAPK. IL-7 plays an essential role in the immune system, particularly in T cell development, survival, and homeostatic proliferation. In?in vitro?cultures, IL-7 supports the survival of na?ve and memory T cells and helps prevent apoptosis. During early B cell development, it promotes both proliferation and differentiation. In the context of cell therapy—such as CAR-T or TCR-T treatments—IL-7 is often used in combination with other cytokines like IL-15 to enhance T cell expansion and maintain stem-like properties. Furthermore, IL-7 can improve the yield and activity of tumor-infiltrating lymphocytes (TILs) or virus-specific T cells during culture.
