- Lymphotoxin-A Alpha (LT-A Alpha), also known as TNF-B Beta, is a member of the TNF superfamily of proteins. NK cells, T cells, B cells, and lymphoid tissue-inducer cells express LT-A Alpha. LT-A Alpha can be secreted as a soluble homotrimer or form membrane bound heterotrimers with lymphotoxin-B Beta (LT A Alpha1B Beta2 or LT A Alpha2B Beta1) which can be cleaved from the cell surface by matrix metalloproteases. Soluble LT-A Alpha binds to and signals through TNFR1/TNFR2, activating the canonical NF-K KappaB pathway. In contrast, LT A Alpha1B Beta2 heterodimers bind to the LTB BetaR receptor and activate the noncanonical NF-K KappaB pathway. As a result, LT-A Alpha and TNF-A Alpha have overlapping functions. Soluble LT-A Alpha and LT A Alpha1B Beta2 play key roles in lymphangiogenesis. The LT A Alpha1B Beta2/LTB BetaR axis is essential for the development of lymphoid tissue.
