- Murine antibodies were produced by immunising BALB/c mice with the mouse erythroleukaemic (MEL) cell line stably expressing high levels of wildtype, full-length human Kell protein. This overexpression allowed several epitopes of Kell protein to serve as the immunogen, resulting in identification of several anti-Kell mAbs with different specificities, of which MIMA-8 targeted the Jsb antigen. Splenocytes were obtained from immunised mice and fused with the P3X63Ag8.653 myeloma cell line to produce a hybridoma.

- MIMA-8 was shown to be specific for the Jsb epitope of the Kell protein using flow cytometry, whereby the mAb agglutinated Jsb+ , but not Jsa+Jsb-, erythrocytes (Chu et al, 1999). These erythrocytes also lacked other high-incidence Kell protein antigens. The Kell protein is a cell-surface metalloprotease, and constitutes one of the major blood antigenic systems of humans.
