Upon binding to its ligands CD80 (B7-1) and CD86 (B7-2)-expressed on antigen-presenting cells (APCs)-CD28 triggers intracellular signaling cascades that promote T cell proliferation, cytokine production, and the survival of activated T cells. Dysregulation of CD28 signaling is linked to autoimmune diseases, immune deficiency, and impaired antitumor immunity, making it a central target for studying immune activation and modulation in murine models.
