The epithelial sodium channel alpha subunit (ENaCA Alpha) is a key component of the heterotrimeric ENaC complex, which regulates sodium ion transport across epithelial membranes. While traditionally studied in renal, pulmonary, and gastrointestinal systems, emerging evidence highlights ENaCA Alpha's relevance in the central nervous system and neurodegenerative disease. ENaCA Alpha modulates ionic homeostasis and fluid balance in neural tissues, contributing to neuroprotection under stress conditions. In models of ocular hypertension, upregulation of ENaCA Alpha in retinal tissues suggests a compensatory mechanism to counteract elevated intraocular pressure, a known risk factor for glaucoma. Additionally, ENaCA Alpha expression is dynamically regulated during keratinocyte differentiation, indicating its broader role in cellular stress responses and tissue remodeling. These findings point to ENaCA Alpha as a potential modulator of neuroinflammation and glial activation-processes central to neurodegenerative pathologies such as Alzheimer's disease and multiple sclerosis. As interest grows in ion channel dysfunction in neurodegeneration, ENaCA Alpha is gaining attention as a novel target for therapeutic intervention. Its role in maintaining ionic equilibrium and responding to mechanical and osmotic stress positions it at the intersection of neuroscience, neuroprotection, and precision medicine.
