- LGI1 (Leucine-Rich Glioma Inactivated 1) is a secreted synaptic protein that modulates neuronal excitability and synaptic transmission. It interacts with ADAM22 and ADAM23 to stabilize AMPA receptor function and maintain synaptic integrity. Mutations in LGI1 are linked to autosomal dominant lateral temporal lobe epilepsy (ADTLE) and have been implicated in autoimmune encephalitis.
