- mGluR5 (metabotropic glutamate receptor 5) is a subtype of glutamate receptor that regulates synaptic transmission and plasticity via G-protein-coupled signaling. It is predominantly expressed in the hippocampus, cortex, and striatum-regions vulnerable to neurodegeneration. Aberrant mGluR5 activity has been linked to excitotoxicity, calcium dysregulation, and neuroinflammation, contributing to the pathophysiology of Alzheimer's disease, Parkinson's disease, and amyotrophic lateral sclerosis (ALS). mGluR5 also interacts with amyloid precursor protein and tau, influencing their aggregation and toxicity.
