- SHANK1 is a postsynaptic density (PSD) scaffolding protein that integrates neurotransmitter receptors and signaling molecules into the cortical cytoskeleton. It plays a key role in synaptic organization, plasticity, and signal transduction at excitatory synapses. SHANK1 recruits proteins such as betaPIX and PAK to dendritic spines, regulating spine morphology and synaptic strength. It also interacts with ionotropic and metabotropic glutamate receptor complexes, contributing to synaptic signaling fidelity.
