P2RY12 encodes the P2Y purinoceptor 12; a G-protein coupled receptor (GPCR) that primarily responds to adenosine diphosphate (ADP). It couples to Gi proteins; leading to inhibition of adenylyl cyclase and reduction of intracellular cAMP levels. P2RY12 is highly expressed on platelets; where it plays an essential role in platelet activation; aggregation; and thrombus formation. It is the pharmacological target of antiplatelet drugs such as clopidogrel; prasugrel; and ticagrelor. Beyond the circulatory system; P2RY12 is also expressed in microglia within the central nervous system; where it regulates microglial chemotaxis and homeostatic responses. Dysregulation of P2RY12 is implicated in thrombosis; stroke; and neuroinflammation.
