- NTSR2 encodes the neurotensin receptor type 2; a G-protein coupled receptor (GPCR) that binds the neuropeptide neurotensin (NTS). It primarily couples to Gq/11 and Gi/o proteins; triggering downstream phospholipase C activation; Ca²⁺ mobilization; and MAPK signaling. NTSR2 is expressed in the central nervous system (notably in the midbrain; hypothalamus; and spinal cord) as well as in peripheral tissues. It mediates neurotensin-induced analgesia; hypothermia; and regulation of dopamine transmission; and contributes to neuronal excitability; pain modulation; and neuroendocrine secretion. Altered NTSR2 expression has been linked to psychiatric disorders; neuroinflammation; and certain cancers; making it a potential therapeutic target in neurobiology and oncology.
