- HTR2A encodes the 5-hydroxytryptamine (serotonin) receptor 2A; a G-protein coupled receptor (GPCR) that mediates many of serotonin’s effects in the central nervous system and peripheral tissues. It primarily couples to Gq/11 proteins; activating the phospholipase C (PLC)–IP₃–Ca²⁺ signaling pathway; leading to neuronal excitation and modulation of neurotransmitter release. HTR2A is highly expressed in the cerebral cortex; hippocampus; and platelets; and regulates mood; perception; sleep; and cognition. Dysregulation or polymorphisms of HTR2A are associated with psychiatric disorders such as schizophrenia; depression; and anxiety; and it is a pharmacological target of antipsychotics; antidepressants; and hallucinogens (e.g.; LSD; psilocybin).
