LifeSensors Targeted Protein Degredation
2BScientific proudly distributes LifeSensors’ plethora of reagents that support multiple stages and applications of Targeted Protein Degradation (TPD) research, ranging from screening to validation. The Ubiquitin Proteasome System (UPS) standardises protein degradation in cells and is a critical target for next generation drug discovery. Technologies such as molecular glues and PROTACs both utilise this system to remove disease-causing proteins and enable researchers to effectively investigate drug effectiveness.
TUBE Embedded Microtiter Plates
TUBEs or “Tandem Ubiquitin Binding Entities” are engineered proteins that bind selectively to ubiquitin chains on a sub-nanomolar level, resulting in artefact free monitoring of ubiquitination events. TUBE assays provide accurate, artefact free data while enabling varied applications ranging from discovering molecular glues and PROTACs with TUBEs, evaluating proteasomal degradation using K48-selective variants, studying autophagy and mitophagy with K63-selective TUBEs, and identifying inflammatory target degraders via M1-linear TUBEs.
Molecular Glue Discovery Tools
Molecular glues promote interactions between E3 ligases and target proteins, supporting selective degradation. They’re simpler than PROTACs but require vital assays for validation. LifeSensors Molecular Glue Plates enable high-throughput screening of molecular glues, allowing natural ubiquitination detection without tags or artificial modifications.
Used alongside LifeSensors’ TUBEs, E3 TR-FRET assays, and PROTAC plates, these powerful tools enable researchers to discover and validate molecular glues, monitor ubiquitination both in vitro and in vivo, and efficiently investigate degradation mechanisms for deeper understanding at a faster throughput.
PROTAC and E3 Ligase Assays
LifeSensors’ E3 ligase assay suite supports validation by directly measuring E3 ligase-mediated ubiquitination through a combination of reagents including E3 TR-FRET assays, E3 ligase substrate assays, and PROTAC plates. These valuable assays provide sensitive, natural readouts to confirm target engagement and degradation.
