Aquaporin 2 (AQP2) is a vasopressin-sensitive water channel primarily expressed in the apical membrane of kidney collecting duct cells. As a member of the aquaporin family of integral membrane proteins, AQP2 facilitates the selective transport of water molecules while excluding ions and solutes, playing a central role in systemic water homeostasis. Under basal conditions, AQP2 resides in intracellular vesicles within renal epithelial cells. Upon stimulation by vasopressin, binding to the V2 receptor activates a cAMP-dependent signaling cascade that triggers the translocation of AQP2-containing vesicles to the plasma membrane, enabling rapid water reabsorption. Mutations or dysfunction in AQP2 are causative of autosomal dominant nephrogenic diabetes insipidus (NDI), a disorder characterized by impaired renal water conservation. Beyond its renal function, AQP2 is gaining attention in neuroscience and neurodegenerative disease research. Vasopressin signaling and water balance are increasingly recognized as modulators of brain function, particularly in the context of neuroendocrine regulation, circadian rhythm, and stress response. Dysregulation of vasopressin-AQP2 pathways has been implicated in conditions such as Alzheimer's disease, where altered fluid homeostasis and blood-brain barrier integrity contribute to disease progression. As research expands into the systemic and central roles of aquaporins, AQP2 emerges as a potential link between peripheral water regulation and central nervous system health, offering new avenues for understanding and targeting neurodegenerative disorders.
