Introduction to DNA Design for myTXTL
Learn the important elements required for the design of DNA templates used in the myTXTL system for either plasmid or linear DNA templates.
myTXTL™ is an E.coli based protein expression system to enable cell free protein transcription translation – an in vitro protein translation and transcription kit (I.V.T.T.)
The advantages of an IVTT kit is the speed at which proteins can be produced – without having to optimise cellular growth conditions and transfection conditions. Some proteins can be difficult to express in live cells, since they can affect cellular metabolism – such as toxic proteins.
With IVTT kits, designed proteins can be made with a synthetic DNA to see if the A.I. predictions are accurate, or bacteriophages can be re-booted, and can eliminate the need for phage screening and subsequent cloning.
As the system is based on bacterial cellular machinery, variations of the myTXTL™ kits include supplements to help protein folding for more complicated mammalian proteins like antibodies.
