MaxLight™490 is a new Blue-Green photostable dye conjugate comparable to DyLight™488, Alexa Fluor™488 and offers better labeling efficiency, brighter imaging and increased immunodetection. Absorbance (491nm); Emission (515nm); Extinction Coefficient 73,000. VEGF (vascular endothelial growth factor) is a homodimeric, disulfide-linked glycoprotein involved in angiogenesis which promotes tumor progression and metastasis. It exhibits potent mitogenic and permeability inducing properties specific for the vascular endothelium. Multiple isoforms of VEGF containing 206-, 189-, 165-, and 121-amino acid residues have been described which arise from alternative splicing of a single gene. The smaller two isoforms, VEGF165 and VEGF121, are secreted proteins and act as diffusible agents, whereas the larger two remain cell associated. VEGF/VPF plays an important role in angiogenesis, which promotes tumor progression and metastasis. Applications: Suitable for use in FLISA and Western Blot. Other applications not tested. Recommended Dilution: Direct FLISA: ≥0.5ug/ml using 100ul/well antibody solution. Allows the detection of 0.2-0.4ng/well of recombinant hVEGF. Western Blot: 0.1-0.2ug/ml. The detection limit for recombinant hVEGF is 1.5-3.0ng/lane, under either reducing or non-reducing conditions. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.  Note: Applications are based on unconjugated antibody.
