- Tyrosine hydroxylase (TH) is the rate-limiting enzyme in the synthesis of the catecholamines dopamine and norepinephrine. TH antibodies can therefore be used as markers for dopaminergic and noradrenergic neurons in a variety of applications including depression, schizophrenia, Parkinson’s disease and drug abuse (Kish et al., 2001; Zhu et al., 2000; Zhu et al., 1999). TH antibodies can also be used to explore basic mechanisms of dopamine and norepinephrine signaling (Witkovsky et al., 2000; Salvatore et al., 2001; Dunkley et al., 2004). Applications: Suitable for use in Western Blot, Immunohistochemistry, Immunofluorescence and Immunocytochemistry. Other applications not tested. Recommended Dilutions: Western Blot: 1:1000; Detects a band of ~60kD in rat caudate cell lysates. Immunohistochemistry: 1:1000 Immunocytochemistry (IF): 1:500-1:1000, fixed with 4% PFA and permeabilized with 0.1-0.3 % Triton X. Optimal dilution determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
