- Trypsin is a member of the serine protease family. Trypsin cleaves peptides on the C-terminal end of lysine and arginine amino acid residues. The optimum pH of trypsin lies between 7-9. The enzyme is inhibited by serine protease inhibitors (e.g. PMSF) and metal chelating agents (e.g. EDTA). Recombinant Porcine Trypsin is a genetically engineered protein that is expressed in E. coli and purified by high pressure liquid chromatography. There is no contaminating enzyme activity from carboxypeptidase A or chymotrypsin. No protease inhibitors such as PMSF are used during the manufacturing process. Source: E. coli (Porcine pancreas) E.C: 3.4.21.4 Appearance: White lyophilized powder Purity: ≥99% (HPLC) Molecular Weight: 23.8kD Protein Content: ≥70% β-trypsin; ≤20% α-trypsin Specific Activity: ≥3800 USP unit/mg protein Unit Definition: One USP unit of trypsin activity will produce a Delta A253 of 0.003 per minute in a reaction volume of 3ml at pH7.6 and 25℃, with BAEE as a substrate (1cm light path). Contaminant Activity: No chymotrypsin, carboxypeptidase A, or other protease contaminants. Storage and Stability: Store in a sealed container at 2-8°C. Once reconstituted, it should be stored at -20°C. Stable for 24 months. Greater than 90% enzymatic activity retains after 10 repeated freeze-and-thaw cycles.
