- Trefoil factor 2 (TFF2), also known as spasmolytic polypeptide (Sp) is a 20kD secreted trefoil factor family protein. It is a protease-resistant glycoprotein primarily produced by mucous-secreting cells of the stomach and duodenum and is upregulated in response to inflammation. TFF2 interacts with mucin and other TFF2-binding proteins to protect and repair mucosal epithelium. TFF2 is also expressed by macrophages and lymphocytes and may have a role in modulating immune system responses. The 106 residue mature human TFF2 contains two trefoil domains (aa # 36-72 and # 80-121) and one N-linked glycosylation site. Human TFF2 shares 71% and 87% aa sequence identity with canine and mouse TFF2, respectively. Applications: Suitable for use in ELISA and Western Blot. Other applications not tested. Recommended Dilution: ELISA (direct): 0.5-1ug/ml. The detection limit for rhTFF2 is ~0.5ng/ml. Western Blot: 0.1-0.2ug/ml. The detection limit for rhTFF2 is ~5ng/ml under non-reducing and reducing conditions. Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
