Extra Details:: Thymidylate Synthase (TS or TYMS or TMS or dTMP synthase) (EC 2.1.1.45) converts deoxyuridine monophosphate (dUMP) to deoxythymidine monophosphate (dTMP for DNA synthesis. TS is a critical target for the fluoropyrimidines, an important group of antineoplastic drugs that are widely used in the treatment of solid tumors. Both 5-FU and fluorodeoxyuridine are converted in tumor cells to FdUMP which inactivates TS by formation of a ternary covalent complex in the presence of the folate cofactor 5,10-methylenetetrahydrofolate. Expression of TS protein is associated with response to 5-fluorouracil (5-FU) in human colorectal, gastric, head and neck, and breast carcinomas. Human TS is a 313aa cytoplasmic, homodimeric enzyme encoded by chromosome 18p11.32. This protein also binds to its mRNA thus repressing its own translation. Applications: Suitable for use in ELISA, Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 1-5ng/lane. Users must optimize antibody dilution depending upon the nature of samples and other technical conditions. TS is ~36kD. 5-FU-resistant cell cancer lines or colon carcinoma or other tumor cells can be used as positive controls. Optimal dilutions to be determined by the researcher. Hybridoma: Sp2/0 splenocytes with spleen cells from Balb/c mice. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile ddH2O or PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
