Extra Details:: Human Thymidylate Synthase and is useful in determining its presence in various assays. Because inhibition of Thymidylate Synthase prevents DNA synthesis and cell proliferation, the enzyme is an important target for cancer chemotherapeutic drugs, specifically the fluoropyrimidine group of antineoplastic drugs used to treat solid tumors. This antibody can detect antigen in a variety of human cells and tissues, as well as bacteria, African green monkey, rat and mouse. Somewhat lower dilutions may be required in some non-human cell lines. Anti- Thymidylate Synthase can detect Thymidylate Synthase by immunochemistry in proliferating cell cultures and tissues but does not stain nonproliferating cells. Normal colon mucosa shows weak staining; however, some colorectal cancer specimens show very strong staining. Applications: Suitable for use in Immunofluorescence, ELISA, Western Blot, Immunoprecipitation and Immunohistochemistry. Other applications not tested. Recommended Dilution: ELISA: 1:10,000 Use HRP Rabbit-anti-Sheep IgG [H&L] and ABTS Stable Liquid Substrate 1.46mM/Liter for HRPO A0030 as a substrate for 30 minutes at RT. Western Blot: 1:10-001:5000 (TMB) Immunohistochemistry (Methanol fixative): 1:100 Immunoprecipitation: 1:500 Pre-clean with a non-specifiic Sheep IgG. Optimal dilutions to be determined by the researcher. *Recommended Secondary Antibodies: I1904-59CIgG, H&L (HRP) Pab RbxSh I1904-59H: IgG, H&L, X-Adsorbed (HRP) Pab RbxSh Storage and Stability: Storage and Stability: May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
