- Extra Details:
- Human T cell receptor (TCR) is a T cell surface expressed heterodimer of a/b or g/d chains. TCR's recognize antigen peptides bound to MHC molecules forming the basis of antigen specific response by T cells. Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilutions: Flow Cytometry: 1:50 (10ug/ml); 80ul of diluted T2040-17C labels 5x10e5 cells. Optimal working dilutions to be determined by researcher. Flow Cytometry Procotol: 1. 5x10e5 cultured CH7C17 tumor cells (HA1.7 Vb3+ transfectant) were washed and incubated 45 minutes on ice with 80ul of T2040-17C at a 1:50 dilution (10ug/ml). 2. Cells were washed three times, fixed and analyzed by FACS. Cells stained positive with a mean shift of 1.54 log10 fluorescent units when compared to a Mouse IgG2a (PE) negative control at a similar concentration. 3. Binding was blocked when cells were pre-incubated with an excess of T2040-17 (unlabeled anti-TCR V beta 3 antibody). Storage and Stability: Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: PE conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap.
