Extra Details:
A sodium-dependent transport system is responsible for transfer and distribution of vitamins (pantothenate, biotin, and ascorbic acid, etc.) to different parts of the body. Since vitamins are required for essential metabolic processes in all mammalian cells, such cells have developed intrinsic mechanisms to achieve accumulation of essential vitamins. Vitamin transporters belong to the Solute Carrier family (SLC) and include Sodium-dependent Multi-Vitamin Transporter (SMVT), Sodium-dependent Vitamin-C Transporter (SVCT) 1 and 2, and Creatine Transporter (CRT1/ CT1). Other vitamin transporters from the SLC family include Thiamine Transporter Protein 1 (THTR1), Folate Transporter or Reduced Folate Carrier 1 (RFC1), Thyroid Iodide Transporter (TIT), Taurine Transporter (TAU) etc. Vitamin C absorption and distribution requires SVCT1 and SVCT2. SVCT2 accounts for tissue specific uptake of Vitamin C. SVCT2 is a 592-amino acid protein in rat (~70kD) and is 650 amino acids long in human (chrm 20, gene SLC23A2). Expression is widespread, occurring in neurons, bone and other tissues. It is the predominant pre-natal Vitamin C transporter to most tissues particularly in the central nervous system and adrenal glands. Deficiency of SVCT2 is lethal in newborn mice. Applications: Suitable for use in ELISA. Western Blot, though not tested, may potentially be used as an application. Other applications not tested. Recommended Dilution: ELISA: 0.5-1ug/ml; Control peptide can be used to coat ELISA plates at 1ug/ml. See S5327-46A. Western Blot: 1-10ug/ml using ECL technique. Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
