- The catalytic domain of serum and glucocorticoid-regulated protein kinase (SGK) is 54% identical with that of Akt/PKB. Although SGK1 lacks the PtdIns(3,4,5) P3-binding Pleckstrin-homology domain, it retains the residues that are phosphorylated by PDK1 (Thr256 and Ser422). PDK1 activates SGK in vitro by phosphorylating Thr256. Like Akt/PKB, SGK1 preferentially phosphorylates serine and threonine residues that lie in RXRXXS/T motifs. SGK1 can be activated by PI-3 Kinase, which makes it an important signaling molecule in growth factor and insulin dependent signaling pathways. Applications: Suitable for use in Western Blot and Immunoprecipitation. Other applications have not been tested. Recommended Dilution: Western Blot: 0.5-2ug/ml detects SGK1 in RIPA lysates from A431 and Rat 2 cells. Rat 2 cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with S1010-83 (0.5ug/ml). Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system. Immunoprecipitation: 4ug Immunoprecipitates SGK1 from 500ug of Rat 2 lysate. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
