- Applications: Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilutions: Western Blot: 1:100 Immunohistochemistry (Bn-SA/HRP Technique): 1:100-1:300 in PBS. Immunohistochemistry (Bn/Av-HRP Technique): 1:500-1:1000 in PBS. Optimal dilutions to be determined by the researcher. Immunohistochemistry: Perfusion Fixation: Fixative-4% paraformaldehyde in 0.1M PBS, pH 7.4; 500ml over ~20 minutes. Post Fixation: 4% paraformaldehyde in 0.1M PBS, pH 7.4; 500ml over ~20min at RT or 1.5hr at 4°C. Note: Paraformaldehyde is a necessary component in fixation of serotonin. If needed, low levels of glutaraldehyde (0.1-0.3%) may be used in conjunction with paraformaldehyde. Sections: 50um vibratome Adsorption control: Synthetic peptide corresponding to aa8-23 of the rat 5-HT7 receptor Incubation on Tissue: 16-24 hours at 4ºC. Positive Control: Rat cortex and hippocampus tissues Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
