- S6 (Ribosomal protein S6, RpS6, 40S ribosomal protein S6) is known to play a role in cell growth and proliferation. It lays downstream of both the PI3K/Akt/mTOR pathway as a substrate of p70 S6 kinase and the Ras/Erk pathway as a substrate of RSK. S6 is phosphorylated in response to stimulation by growth factors, tumor promoting agents, and mitogens. It is dephosphorylated at growth arrest. There are two major areas of S6 phosphorylation; Ser235/Ser236 that is known to be phosphorylated by both p70S6 Kinase as well as RSK as well as Ser240/244 which is thought to only be phosphorylated by p70S6K. Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 1:500-1:2000 Optimal dilutions to be determined by the researcher. Positive Control: Serum stimulated 3T3 cell lysate. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
